Abstract: Triterpene glycoside in term of asiaticoside was extracted from Centella asiatica sp. through Enzyme assisted extraction (EAE) method. Triterpene glycoside is a secondary metabolite in Centella asiatica that can heal skin wounds with its anti-inflammatory properties. The addition of cellulase enzyme increases the leaching ability of the solvent used in this extraction, which is ethanol, by breaking up the plant's cell walls. The extraction yield of the triterpenoid compound, asiaticoside was determined using High-performance liquid chromatography (HPLC). Several parameters were studied such as different volumes of enzyme (0.1 mL, 0.2 mL, and 0.3 mL), extraction time (40 min, 60 min and 80 min) and solvent concentration (30%, 50%, 70% and 90%) (v/v) of ethanol. Phytochemical screening using Salkowski’s test showed the formation of reddish-brown color, indicating the presence of glycosides for all samples. HPLC result showed that the highest yield of asiaticoside through EAE method was 0.1239 mg/mL under condition of 0.2 mL of enzyme, 80 min of extraction time and 70% of solvent concentration. The Scanning Electron Microscopy (SEM) shows a massive difference in the physical change of surface structure of the sample before and after the extraction. In conclusion, from the findings of this study, EAE showed a high potential for extracting triterpene glycosides while being a green and eco-friendly extraction method.
